摘要: Abstract Hepatocytes were isolated from male Sprague-Dawley rats 30 min after oral challenge with carbon tetrachloride (CCl 4 ), or 1 hr ip injection of d (+)-galactosamine (GAL). Cell preparations comparable yield and initial viability obtained toxin-treated respective control animals equivalent 1-hr attachment efficiencies to tissue culture plastic. Cells exhibited significant degeneration over 48 in culture. This included loss viable cell density on the monolayer increased lactate dehydrogenase activity media compared controls. Toxicity expressed was dose dependent for both CCl (0.1–2.5 ml/kg) GAL (100–400 mg/kg). Loss vitro ultrastructural followed a time course consistent hepatocellular necrosis produced by these agents vivo . As model studying late occurring events progression initiation toxic injury death, this methodology offers several potential advantages strict models. The analytical an are incorporated physiologically relevant doses hepatotoxins. Limited bioactivating hepatocytes, limitations suspension hepatocytes toxicity studies, also circumvented model.
