Isolation and maintenance of monolayer hepatocytes from the livers of toxin-treated rats

摘要: Abstract Hepatocytes were isolated from male Sprague-Dawley rats 30 min after oral challenge with carbon tetrachloride (CCl 4 ), or 1 hr ip injection of d (+)-galactosamine (GAL). Cell preparations comparable yield and initial viability obtained toxin-treated respective control animals equivalent 1-hr attachment efficiencies to tissue culture plastic. Cells exhibited significant degeneration over 48 in culture. This included loss viable cell density on the monolayer increased lactate dehydrogenase activity media compared controls. Toxicity expressed was dose dependent for both CCl (0.1–2.5 ml/kg) GAL (100–400 mg/kg).
