摘要: A population of Saccharomyces cerevisiae was cultured for approximately 450 generations in the presence high glucose to select genetic variants that grew optimally under these conditions. Using parental strain BY4741 as starting population, an evolved culture obtained after aerobic growth a medium generations. After evolution period, three single colony isolates were selected analysis. Next-generation Ion Torrent sequencing used evaluate changes. Greater than 100 deletion/insertion changes found with half effecting genes. Additionally, over 180 SNPs identified more one-quarter resulting nonsynonymous mutation. Affymetrix DNA microarrays and RNseq analysis determine differences gene expression strains compared strain. It established 900 genes demonstrated significantly altered relative Many showed similar alterations their all strains. Interestingly, included role oxidative metabolism. Overall results are consistent physiological observations optimal carbon source. Namely, decreased ethanol production suggest underlying metabolism switched from fermentation respiration during selection on glucose.
