摘要: Publisher Summary The free-living, unicellular, ciliated protozoan Tetrahymena has been widely used for diverse biochemical investigations because of ease in growing cells large quantities media that are axenic or even completely defined. In addition, tetrahymena offers various other advantages—namely, (1) well-defined development subcellular organelles, (2) strikingly different distribution specific lipids among the functionally distinct membranes within cell, and (3) sensitive adjustment to environmental factors such as starvation, temperature, chemicals. Various membrane components include nuclei, mitochondria, lysosomes, endoplasmic reticulum, cilia, pellicles, oral apparatus, food vacuoles. This chapter reviews procedures isolation from pyriformis. methods divided into two groups: systematic by which several can be isolated same batch culture, nonsystematic individual organelles separately isolated. For studies requiring at metabolic level, method must employed.
