摘要: Abstract Schisandrin, derived from the Chinese medicinal herb Schisandra chinensis, has been found to confer protective effects on circulation systems. But underlying molecular mechanisms remain unclear. The aim of this study was investigate a high level glucose RhoA and eNOS activity in human umbilical vein endothelial cells(HUVECs) how Schisandrin plays role mediating these effects. To find optimal treatment time, HUVECs were cultured at concentration (30 mM) for different lengths time (0, 12, 24, 48 h). Subsequently, cells randomized into five groups: normal group, three groups that given doses (5, 10, 20 μM) Schisandrin. pretreated with 24 h before stimulation glucose. morphology various assessed under light microscope. Immunocytochemical staining used detect p-MYPT1 expression. levels determined using Activation Assay Biochem Kit. examined nitrate reduction test. results showed increased reduced, thus decreasing secretion NO. However, after pretreatment (10, 20 μM), inhibited increased, which led an increase NO production compared group.
