Comparative Proteomic Analysis of the Hepatic Response to Heat Stress in Muscovy and Pekin Ducks: Insight into Thermal Tolerance Related to Energy Metabolism

摘要: The Pekin duck, bred from the mallard (Anas platyrhynchos) in china, is one of most famous meat duck species world. However, it more sensitive to heat stress than Muscovy which believed have originated South America. With temperature raising, mortality, laying performance, and quality are severely affected. This study aims uncover temperature-dependent proteins two using comparative proteomic approach. Duck was cultured under 39°C ± 0.5°C for 1 h, then immediately returned 20°C a 3 h recovery period, liver were extracted electrophoresed two-dimensional mode. After analysis gel images, 61 differentially expressed detected, 54 clearly identified by MALDI TOF/TOF MS. Of protein spots identified, 7 found both species, whereas 47 specific (25 22 duck). As well known, chaperone proteins, such as shock (HSP) 70 HSP10, abundantly up-regulated response stress. we also that several α-enolase, S-adenosylmethionine synthetase, showed different expression patterns 2 species. enriched biological processes grouped into main categories according gene ontology analysis: cell death apoptosis (20.93%), amino acid metabolism (13.95%) oxidation reduction (20.93%). mRNA levels investigated real-time RT-PCR. To our knowledge, this first provide insights differential following ducks enables better understanding possible mechanisms animals.
