Effects of retinal glial cells on isolated rat retinal ganglion cells.

摘要: PURPOSE. The effect of retinal glial cells on ganglion cell (RGC) survival was investigated in cocultures pure, isolated with RGCs. METHODS. RGCs from 2-day-old rats were cocultured for 48 hours, avoiding direct contact between types, either nonconfluent 3-day-old or confluent 3-day-old, 12-day-old, 1-year-old rats. Survival evaluated by flow cytometry. Amino acids determined culture medium. effects glutamate antagonists, 6-cyano-7-nitroquinoxaline-2,3-dione and MK801, a nitric oxide (NO) scavenger, 2-(4-carboxyphenyl)-4,4,5,5tetramethylimidazoline-1-oxyl-3-oxide potassium salt (c-PTIO), an NO synthase inhibitor, N G -nitro-L-arginine methyl ester (L-NAME), examined. RESULTS. Nonconfluent significantly reduced the small large RGCs, but only regardless rat's age at time harvesting. Profiles some amino varied, depending condition. Cocultures released more into medium than pure culture. antagonists improved cells, especially when two administered combination, case c-PTIO L-NAME, also cells. CONCLUSIONS. Adverse varied size confluence. Glutamate may be involved cell-related antisurvival effects.
