摘要: The structural changes which occur in chromatin DNA after ischemic brain injury are poorly understood. This study examined the appearance of double-strand breaks and temporal profile degradation following focal rat brain. Focal cerebral ischemia was produced by tandem occlusion common carotid proximal middle arteries. effects decapitation were also studied analysis. extracted standard methods from tissues electrophoresed on a 1.5% agarose gel. With ischemia, random cleavage observed as dense "smear" gel electrophoresis beginning 6 h insult, increasing amount thereafter. provided fragmentation, is specific at internucleosomal linker regions, particularly caudoputamen. To determine whether an endonuclease responsible for present, nuclear proteins normal nuclei activity determined using plasmid incubation system. demonstrated that have Ca(2+)-dependent related to fragmentation. Ischemic causes both brain, probably mediated endonuclease.
