摘要: Background 'Bianliang ziyu', a famous chrysanthemum variety commonly planted in Kaifeng, China, is often consumed by local residents. However, the hepatoprotective effects of Bianliang ziyu and their underlying mechanisms are not clear. Objective In this study, we investigated antioxidative extract (BZE) on liver injury explored its molecular mechanisms. Design Sprague-Dawley rats were administered BZE intragastric administration for 8-9 days, then alcohol or carbon tetrachloride (CCl4) was gavage to induce acute injury. The activities serum alanine aminotransferase, aspartate superoxide dismutase, malondialdehyde measured, each rat examined histopathological changes. vitro, HL-7702 cells pretreated with 24 h exposed 30 mmol•L-1 acetaminophen (APAP) 12 h. survival rate aminotransferase determined. Then, oxidative stress, apoptosis, activation nuclear factor erythroid-2-related 2 (Nrf2) signaling induced APAP. Results results showed that prevented alcohol-, CCl4-, APAP-induced suppressed hepatic stress vitro vivo. also observed significantly inhibit reduction mitochondrial membrane potential regulate expression Bcl-2, Bax Caspase-3 cells. addition, promoted translocation Nrf2 as well downstream gene hemeoxygenase-1 (HO-1) vitro. Furthermore, findings siRNA reversed cell apoptosis-related protein Conclusions plays an important role preventing hepatotoxicity inhibiting apoptosis through signaling. could be developed effective functional food protecting liver.
