The GlxR regulon of the amino acid producer Corynebacterium glutamicum: Detection of the corynebacterial core regulon and integration into the transcriptional regulatory network model

摘要: Corynebacterium glutamicum is an industrially important producer of amino acids and emerging model system for the Corynebacterineae. The glxR gene C. ATCC 13032 encodes a DNA binding transcription factor Crp-Fnr protein family. Available data indicated prominent role GlxR in transcriptional regulatory network glutamicum. We have used recently published whole-genome annotations several corynebacteria to derive further evidence GlxR-mediated regulation from comparative genomics approach. A bioinformatic strategy detected 22 genes belonging proposed core regulon corynebacteria. Binding purified 40-mer oligonucleotides representing predicted sites upstream region was verified vitro by electrophoretic mobility shift assays. Based on current data, reliable genome-scale prediction performed, indicating that about 14% annotated might be under direct control GlxR. Integration interactions with stored CoryneRegNet enabled reconstruction refined This exhibited hierarchical modular structure characterized presence master regulators each functional module, serving as dominating hub.
