摘要: AIM: To investigate the effect of oridonin on nuclear transcription factors and to study relationship between biological behavior inflammatory in human pancreatic cancer (BxPC-3) cells. METHODS: BxPC-3 cells were treated with various concentrations oridonin, viability curves generated test for inhibitory effects drug cells. The expression cytokines such as interleukin-1β (IL-1β), IL-6, or IL-33 was detected cell supernatants using an enzyme-linked immunosorbent assay (ELISA), protein including factor κB, activating protein-1, signal transducer activator 3, bone morphogenetic 2, transforming growth β1 sma mad homologues Western blot. Carcinoma hallmark-related proteins survivin, vascular endothelial factor, matrix metallopeptidase 2 also immunoblotting, intra-nuclear immunofluorescent staining. RESULTS: Treatment reduced a dose dependent manner. exhibited following treatment 8 μg/mL (13.05% ± 3.21%, P < 0.01), highest ratio 90.64% 0.70%, which achieved at 32 μg/mL. IC50 value 19.32 ELISA analysis revealed that down-regulated IL-1β, dose-dependent IL-1β significantly 16 groups compared control group (12.97 0.45 pg/mL, 11.17 0.63 pg/mL vs 14.40 0.38 0.01). Similar trends observed IL-6 expression, (4.05 0.14 4.45 0.43 0.05; 3.95 0.13 8, 16, (911.05 14.18 945.25 12.09 802.70 11.88 768.54 10.98 blot staining analyses suggested changed hallmarks regulated factors. CONCLUSION: results obtained suggest alters through regulation factors.
