摘要: Stem cell exhaustion plays a major role in the ageing of different tissues. Similarly, vitro during expansion prior to their use regenerative medicine can severely compromise stem quality through progressive declines differentiation and growth capacity. We utilized non-destructive multispectral assessment native autofluorescence investigate metabolic mechanisms mesenchymal (MSC) human bone marrow MSCs over serial passages (P2-P10). The spectral signals for NAD(P)H, flavins protein-bound NAD(P)H were successfully isolated using Robust Dependent Component Analysis (RoDECA). decreased course hMSC absolute terms as well relative (optical redox ratio). Relative changes other fluorophore levels (flavins, NAD(P)H) suggested that this reduction was due nicotinamide adenine dinucleotide depletion rather than shift from glycolysis oxidative phosphorylation. Using features, which are determined without fixation or fluorescent labelling, we developed externally validated reliable, linear model could accurately categorize age culture-expanded hMSCs. largest characteristics occurs early ageing. These findings demonstrate feasibility applying technology non-invasive monitoring MSC health vitro.
