Characterization of isolated villus and crypt cells from the small intestine of the adult mouse

摘要: Suspensions of sequentially isolated villus and crypt cells were obtained in order to study certain biochemical changes associated with differentiation epithelial the small intestine mouse. Microscopic observation various cell fractions reveals that detach as individual or sheets epithelium from tip base villus, whereas regions are separated entire units. The retain their ultrastructural integrity judged by electron microscopy. Chemical characterization shows total cellular protein content, expressed activity per cell, remains relatively constant throughout region followed a noticeable drop zone. On other hand, sharp variations values DNA content observed zone depending on reference being used. Activity profiles several brush border enzymes confirm occur during migration tip, species, maximum sucrase mid-villus region, glucoamylase, trehalase, lactase maltase upper third alkaline phosphatase at tip. Forty-eight-hour suspension cultures corresponding zones show moderate decrease enzyme activities approximately 70% original value, remaining stable incubation period. use indicators culture is discussed.
