摘要: PURPOSE Considering tumor-induced suppression of natural killer (NK) cell activity the aim this study was to investigate in vitro effect a standard immunotherapeutic cytokine, interferon (IFN)α, and less investigated agent, 13-cis retinoic acid (RA) on functional receptor characteristics CD16-defined NK cells their functionally diverse dim bright subsets patients with metastatic melanoma (MM). METHODS Peripheral blood lymphocytes (PBL) clinical stage IV MM were stimulated for 18 h RPMI 1640 culture medium (CM) alone, CM supplemented IFN-α (250 U7sol;ml), RA (10-6M) combination. determined using 4 radioactive cytotoxicity assay, while expression activating (NKG2D, CD1617rpar; inhibitory (CD158a, CD158b) receptors CD3-CD16+ analyzed by flow cytometry. RESULTS cytotoxic increased after treatment alone combination RA, only induced increase NKG2D CD161 expression. Contrary this, KIR CD158b. IFN-α-obtained due its induction both subsets, CD16bright subset. CONCLUSION The favorable enhancement obtained is associated upregulation receptors, lack RA-associated probably shown CD158b agent.
