NAD(P)H utilization in the reduction of pyruvate to lactate in a glycogen-containing subline of Ehrlich ascites tumour cells

摘要: The possible pathways of utilization glucose-6-phosphate (G-6-P) produced from glycogen breakdown have been investigated in a glycogen-containing subline Ehrlich ascites tumour cells. Addition either mitochondrial inhibitors or pyruvate to cells metabolizing endogenous substrates enhances the rate lactate production. However, only former condition such effect is abolished by iodoacetate (IAA). In pyruvate-supplemented cause further increase production which becomes insensitive IAA when are depleted substrates. Measurements content show that presence there stimulation glycogenolysis. Significant changes (about 10–20 fold increase) G-6-P level observed both and IAA, irrespective addition. with accumulation lower if starved. results obtained indicate our conditions during glycogenolysis may be oxidized through Embden-Meyerhof pathway phosphogluconate pathway. Indeed, whereas promote this metabolite first route enhancing activity phosphofructokinase, added favours other lowering cytosolic NADPH/NADP+ ratio.
