MANF regulates dopaminergic neuron development in larval zebrafish.

摘要: Mesencephalic astrocyte derived neurotrophic factor (MANF) is recognized as a dopaminergic factor, which can protect neurons from neurotoxic damage. However, little known about the function of MANF during vertebrate development. Here, we report that expression widespread embryonic development and in adult organs analyzed by qPCR situ hybridization zebrafish. Knockdown with antisense splice-blocking morpholino oligonucleotides resulted no apparent abnormal phenotype. Nevertheless, dopamine level morphants was lower than wild type larvae, levels two tyrosine hydroxylase gene transcripts were decreased decrease neuron number certain groups th1 th2 cells diencephalon region observed. These defects rescued injection exogenous manf mRNA. Strikingly, mRNA could partly restore positive Nr4a2-deficient larvae. results suggest involved regulation system
