Analysing calcium signalling of cells under high shear flows using discontinuous dielectrophoresis

摘要: Immobilisation of cells is an important feature many cellular assays, as it enables the physical/chemical stimulation cells; whilst, monitoring processes using microscopic techniques. Current approaches for immobilising cells, however, are hampered by time-consuming processes, need specific antibodies or coatings, and adverse effects on cell integrity. Here, we present a dielectrophoresis-based approach robust immobilisation analysis their responses under high shear flows. This quick label-free, more importantly, minimises electric field integrity, activating short duration 120 s, just long enough to immobilise after which culture media (such HEPES) flushed through platform. In optimal conditions, at least 90% remained stably immobilised, when exposed stress 63 dyn/cm 2 . was used examine shear-induced calcium signalling HEK-293 expressing mechanosensitive ion channel, transient receptor potential vaniloid type 4 (TRPV4), full physiological range stress.
