摘要: The connexin 26 (Cx26) gene suppresses the growth of HeLa cells in vitro and vivo. We explored possibility that Cx26 not only but can also mediate bystander effect is observed some therapy. In therapy mediated by herpes simplex virus thymidine kinase, toxicity ganciclovir affects transduced with neighboring tumor cells; it has been suggested gap junctional intercellular communication (GJIC) may play a role such effect. expressing (Cx26+) or were transfected kinase (tk+) gene, producing Cx26(-)-tk-, Cx26(-)-tk+, Cx26+-tk-, Cx26+-tk+ cells. By making different kinds cocultures these cells, we clear killing effect, assessed neutral red test, coculture Cx26+-tk-/Cx26+-tk+ was markedly prevented long-term inhibitor GJIC, 18-alpha-glycyrrhetinic acid, demonstrating major part seen occurred through Cx-mediated GJIC. These data suggest using Cxs as both suppressor genes diffusers therapeutic approaches.
