摘要: Systematic experiments have resulted in a method of decalcification which, although specifically developed for cytological study reptilian aural tissue, should be applicable to other problems involving electron microscopy organic components bone or tissues closely associated with bone. Reptilian otic capsules and control specimens mammalian hepatic tissue were subjected planned variations fixatives decalcifying agents, embedded, sectioned, evaluated by light microscopy. Additionally, applicability the more satisfactory agents was checked limited testing on from Mongolian gerbils. Acceptable results achieved fixed glutaraldehyde, then decalcified cold 0.1 M solutions disodium tetrasodium EDTA (adjusted pH 7.2–7.4 sodium hydroxide versenic acid, respectively), containing 4.0% glutaraldehyde; following decalcification, washed buffer, postfixed osmium tetroxide, embedded epoxy resins minor modification standard techniques. detail morphological relationships retained, extended treatment solution did not apparently result degradation tissues.
