摘要: Previous studies have suggested the persistence of oligodendrocyte progenitor cells in adult mammalian subcortical white matter. To identify progenitors human matter, we transfected dissociates capsular matter with plasmid DNA bearing gene for green fluorescence protein (hGFP), placed under control early promoter (P2) oligodendrocytic cyclic nucleotide phosphodiesterase (P/hCNP2). Within 4 d after transfection P/hCNP2:hGFP, a discrete population small, bipolar were noted to express GFP. These A2B5-positive (A2B5+), incorporated bromodeoxyuridine vitro , and constituted <0.5% all cells. Using fluorescence-activated cell sorting (FACS), P/hCNP2-driven GFP+ then isolated enriched near-purity. In weeks FACS, most P/hCNP2:hGFP-sorted matured as morphologically antigenically characteristic oligodendrocytes. Thus, harbors mitotically competent cells, which give rise primarily oligodendrocytes . By using fluorescent transgenes GFP expressed an promoter, these may be extracted purified from sufficient numbers implantation cell-based therapy.
