Human Bone Marrow-Derived Mesenchymal Stromal Cell-Seeded Bone Biomaterial Directs Fast and Superior Mandibular Bone Augmentation in Rats

摘要: Atrophic maxillary ridges present a challenge in the field of oral implantology. Autologous bone is still considered gold standard grafting material, but increased morbidity and surgical complications represent major drawback for its use. The aim this study was to assess efficacy an off-the-shelf cell-seeded biomaterial mandibular augmentation, compared acellular counterpart. We used rat model test osteogenic properties marrow-derived mesenchymal stromal cells (MSCs)-seeded microparticles alone. Rats were euthanized at 4 8 weeks, results analyzed using micro-CT imaging, histology (H&E, Masson’s Trichrome), histomorphometry immunohistology (Tartrate-Resistant Acid Phosphatase-TRAP, Osteocalcin human specific anti-mitochondria antibodies). Micro-CT analysis demonstrated that achieved significantly more volume formation weeks (22.75 ± 2.25 mm3 vs 12.34 ± 2.91 mm3, p = 0.016) (64.95 ± 5.41 mm3 42.73 ± 10.58 mm3, p = 0.029), microparticles. Histology confirmed almost completely substituted opposition group. Immunohistochemical showed higher number TRAP positive group group, thereby demonstrating rate remodeling presence MSCs. grafted remained viable detected up least as observed antibody. This material available unlimited quantities could therefore significant advance augmentation by providing larger new shorter time.
