摘要: During DNA synthesis, most polymerases and reverse transcriptases select against ribonucleotides via a steric clash between the ribose 2'-hydroxyl group bulky side chain of an active-site residue. In this study, we demonstrated that human polymerase lambda used novel sugar selection mechanism to discriminate ribonucleotides, whereby was excluded mostly by backbone segment slightly Y505. Such further be dependent on size orientation substituent covalently attached at ribonucleotide C2'-position.
