Genotyping of Multiple Clinical Samples with a Combined Direct PCR and Magnetic Lateral Flow Assay.

摘要: Summary Developing a sensitive, low-cost, and easy-to-use point-of-care testing system for genotyping is important informing treatment decisions predicting the risk of underlying diseases. Conventional methods normally require complex operational procedures as well expensive sophisticated instruments. Here, we report general approach that enables us to detect genotype multiple sample types directly without DNA purification. Moreover, PCR results can be further quantitatively analyzed based on magnetic lateral flow assay (MLFA) system, which avoids steps needed conventional nucleic acid biosensors. As demonstration, show three genotypes aldehyde dehydrogenase 2 (ALDH2) identified using small volume with an accuracy 100% sensitivity 1.0 × 102 cells/μL, are better than those gold standard methods. We believe direct PCR-MLFA represents significant advance toward development portable, sensitive biomedical platforms.
