摘要: In recent years, the discovery of novel nucleic acid-based drug candidates (e.g., siRNA and miRNA) groundbreaking studies for somatic cell reprogramming into a state pluripotency have led to reconsideration use human gene therapy as new paradigm with great therapeutic potential. However, success is dependent on overcoming intra- extracellular barriers hampering efficient delivery acid therapeutics target cells or tissues. Despite relatively low transfection efficiency, attention has been directed cationic polymers dendrimers due their ability condense DNA RNA molecules nano-sized particles which necessary prerequisite transfer acids cells. These carriers show remarkable adaptability significant capacity larger sizes materials. Polyamidoamine (PAMAM) dendrimer employed non-viral carrier its globular shape well-defined structure containing abundant amino surface groups provide possibility decoration via conjugation various moieties. this review, we brought out functionalization strategies PAMAM amines using different pendant moieties such acids, proteins, cyclodextrins, hydrophobic units in order overcome barriers. surface-decorated possessing favorable properties substantial information insight redesigning existing polymers. By understanding role played by conjugated moieties, more designs vehicles may be possible.
