Efficient routing of glucocerebrosidase to lysosomes requires complex oligosaccharide chain formation

摘要: The biosynthesis and intracellular transport of the membrane-associated lysosomal enzyme glucocerebrosidase was studied in monoblast cell line U937. Addition to cultures oligosaccharide trimming inhibitors swainsonine or deoxymannojirimycin led an increased activity glucocerebrosidase. This due prevention degradation enzyme. When homogenates control cells were fractionated on Percoll gradients glucocerebrosidase, like beta-hexosaminidase, distributed two peaks, one at low density high density. cultured presence beta-hexosaminidase still peaks but found mainly fractions also containing galactosyltransferase activity. It is concluded that complex type chain formation required for efficient routing lysosomes U937 cells.
