摘要: Ligating FcγR on macrophages results in suppression of IL-12 production. We show that ligation selectively down-regulates p40 and p35 gene expression at the level transcription. The region responsive to this inhibition maps Ets site promoter. PU.1, IFN consensus sequence binding protein, c- Rel form a complex element upon macrophage activation. Receptor abolishes PU.1-containing activation complex, abrogates A dominant-negative construct PU.1 diminishes promoter activity endogenous protein secretion. Thus, specificity down-regulation following receptor lies These findings provide evidence demonstrating for first time importance transcriptional regulation expression.
