Identification of a novel cytochrome P450 CYP3356A1 linked with insecticide detoxification in Bradysia odoriphaga.

摘要: BACKGROUND Cytochrome P450 monooxygenases play an important role in the metabolic detoxification of insecticides insect pests. However, little is known about a specific gene and its responses to insecticide exposure Bradysia odoriphaga, major pest Chinese chive production. RESULTS In this study, novel gene, CYP3356A1, was cloned from odoriphaga. The full-length cDNA sequence CYP3356A1 2153 bp open reading frame (ORF) encodes 508 amino acids. Quantitative real time PCR(qRT-PCR) analyses different tissues showed that expression highest Malpighian tubule. Moreover, among developmental stages insect, found fourth-instar larvae. Expression upregulated by treatment with imidacloprid, thiamethoxam, β-cypermethrin at median lethal concentrations (LC50 ). RNA interference (RNAi)-mediated silencing significantly increased mortality 36.90%, 25.17%, 36.73 when B. odoriphaga larvae were exposed β-cypermethrin, respectively, LC50 dose. CONCLUSION These results demonstrate related study also our understanding molecular mechanisms insect. © 2018 Society Chemical Industry.
