Mycoplasma contamination in human leukemia cell lines. II. Elimination with various antibiotics.

摘要: 19 suspension cell lines were treated with antibiotics for elimination of chronic contamination mycoplasma. We compared the efficiency, cytotoxicity and cross-resistance commercially available antiboitics MRA (Mycoplasma Removal Agent, a quinolone derivative DNA gyrase inhibitor), Ciprobay (ciprofloxacin, also gyrade BM-cyclin (a combination tiamulin, pleuromutilin derivative, minocycline, tetracycline both inhibitors protein synthesis on ribosomes). Contaminants eliminated in all by BM-Cyclin. Only 74% cleared Ciprobay. Successful treatment was monitored three mycoplasma detection assays. Cross-resistance noted between four five not either reagent. This resistance could, however, be overcome consecutive exposure to BM-cyclin. Employed at recommended concentrations, did cause marked cytotoxicity, but growth cells affected various degrees some antibiotics. The from chronically contaminated is an effective alternative other protocols, cost-intensive time-consuming; lasting damaging effects treatments eukaryotic cannot excluded. Long-term post-treatment monitoring mandatory, since contaminants may only suppressed then recur.
