摘要: The blood–brain barrier (BBB) plays an important role in controlling the passage of molecules from blood to brain extracellular fluid. multidrug efflux pump P-glycoprotein (P-gp) is highly expressed luminal membrane endothelium and contributes formation a functional lipid-soluble drugs such as anticancer agents. mdr1a P-gp-encoding gene exclusively rodent BBB. Primary cultures rat endothelial cells GP8.3 showed dramatic decrease mRNA level some expression mdr1b mRNA. GPNT cells, derived after transfection with puromycin resistance gene, were chronically treated 5 µg/mL puromycin, P-gp substrate. Compared exhibited very high together moderate expression. Accordingly, activity strongly increased. When puromycin-starved was selectively High may thus be related chronic treatment puromycin. We conclude that used valuable vitro model for studying regulation at BBB level.
