Quantification of microRNAs directly from body fluids using a base-stacking isothermal amplification method in a point-of-care device.

摘要: MicroRNAs have been proposed to be a class of biomarkers disease as expression levels are significantly altered in various tissues and body fluids when compared healthy controls. As such, the detection quantification microRNAs is imperative. While many methods established for microRNAs, they typically rely on time consuming handling such RNA extraction, purification, or ligation. Here we describe novel method using direct amplification without upstream sample preparation. Tested with point-of-care device (termed Gene-Z), presence microRNA promotes base-stacking hybridization, subsequent between two universal strands. The approach, which was achieved <60 min, provided sensitivity 1.4 fmol per reaction. percentages whole blood, plasma, faeces, precision (coefficient variation = 2.6%) maintained comparable pristine samples. Overall, developed represents significant step towards rapid, one-step microRNAs.
