摘要: Tail regeneration in Xenopus tadpoles is a favorable model system to understand the molecular and cellular basis of tissue regeneration. Although turnover extracellular matrix (ECM) key event during injury repair, no functional studies evaluate its role appendage have been performed. Studying Hyaluronan (HA), an ECM component, particularly attractive because it can activate intracellular signaling cascades after injury. Here we studied function HA components pathway tadpole tail We found that transcripts for this pathway, including synthase2 ( HAS2 ), Hyaluronidase2 receptors CD44 RHAMM , were transiently upregulated regenerative bud amputation. Concomitantly, increase levels was observed. Functional experiments using 4-methylumbelliferone, specific HAS inhibitor blocked amputation, transgenesis demonstrated required early phases Proper are sustain proliferation mesenchymal cells bud. Pharmacological genetic inhibition GSK3β sufficient rescue when synthesis blocked, suggesting downstream pathway. component cell In addition, crosstalk between demonstrated.
