摘要: Ducks and wild waterfowl perpetuate all strains of influenza viruses in nature. In their natural host, typically cause asymptomatic infection little pathology. are often resistant to capable killing chickens. Here, we show that the virus sensor, RIG-I, is present ducks plays a role clearing an infection. We evidence suggesting RIG-I may be absent chickens, providing plausible explanation for increased susceptibility compared with ducks. detects RNA ligands derived from uncapped viral transcripts initiates IFN response. this study, chicken embryonic fibroblast cell line, DF-1, cannot respond ligand. However, transfection duck into DF-1 cells rescues detection ligand induces IFN-β promoter activity. Additionally, expressing have augmented response resulting decreased replication after challenge either low or highly pathogenic avian virus. Implicating antiviral vivo, found expression induced 200 fold, early innate immune challenged H5N1 A/Vietnam/1203/04. Finding disease resistance gene opens possibility increasing through creation transgenic chicken.
