摘要: 1,25-Dihydroxyvitamin D3 (1,25-(OH)2D3) inhibits the proliferation of mitogen-stimulated human mononuclear cells (MNC) as well production a number proinflammatory cytokines, including interleukin (IL)-1 alpha, IL-6, tumour necrosis factor-alpha, IL-2, interferon-gamma (IFNg) and lymphotoxin (LT). These effects are most likely mediated via specific vitamin D receptors expressed by monocytes activated T lymphocytes. In present study we have evaluated ability 1,25-(OH)2D3 to affect cytokine cell lines stimulated anti-CD3 antibodies or plus anti-CD28 antibodies. selectively reduced supernatant levels while IFNg LT were unaffected. This was followed time- dose-dependent reduction in proliferation. Although expression high affinity IL-2 (IL-2R) (p75) unaffected, exogenously added failed restore The demonstrates that lines, absence accessory cells, may be direct target for 1,25-(OH)2D3, resulting inhibition mechanism which involves interference with activation signals at IL-2R level post level.
