摘要: The high-mannose patch on HIV Env is a preferred target for broadly neutralizing antibodies (bnAbs), but to date, no vaccination regimen has elicited bnAbs against this region. Here, we present the development of bnAb lineage targeting in an HIV-1 subtype-C-infected donor from sub-Saharan Africa. Abs first acquired autologous neutralization, then gradually matured achieve breadth. One Ab neutralized >47% strains with only ∼11% somatic hypermutation and insertions or deletions. By sequencing env, determined key residues that triggered participated Ab-Env coevolution. Next-generation repertoire showed early expansive diversification followed by independent maturation individual limbs, several them developing notable breadth potency. Overall, findings are encouraging vaccine standpoint suggest immunization strategies mimicking evolution entire promoting multiple diverse pathways.
