Response of Cultured Maize Cells to (+)-Abscisic Acid, (-)-Abscisic Acid, and Their Metabolites

摘要: The metabolism and effects of (+)-S- (-)-R-abscisic acid (ABA) some metabolites were studied in maize (Zea mays L. cv Black Mexican Sweet) suspension-cultured cells. Time-course studies metabolite formation performed both cells medium via analytical high-performance liquid chromatography. Metabolites isolated identified using physical chemical methods. At 10 [mu]M concentration 28[deg] C, (+)-ABA was metabolized within 24 h, yielding natural (-)-phaseic [(-)-PA] as the major product. unnatural enantiomer (-)-ABA less than 50% h gave primarily (-)-7[prime]-hydroxyABA [(-)-7[prime]-HOABA], together with (+)-PA ABA glucose ester. distribution different, reflecting different sites membrane permeabilities conjugated nonconjugated metabolites. results imply that oxidized to (-)-PA inside cell, whereas converted (-)-7[prime]-HOABA at cell surface. Growth inhibited by (+)- (-)-ABA, only weak contributions from their caused a inhibition growth approximately 1 [mu]M, for its 50 [mu]M. active (+)-ABA, observed about weakly active, 500 pH indicated transient increase (+0.3 units) 6 after addition not controls or samples treated (-)-PA. effect on Ph marginal. No racemization C-1[prime] during studies.
