摘要: This study used a culture-independent molecular approach to investigate the archaeal community composition of thermophilic bioleaching reactors. Two culture samples, MTC-A and MTC-B, grown with different concentrations chalcopyrite (CuFeS2), copper sulfidic ore, at temperature 78 degrees C pH 1.6 were studied. Phylogenetic analysis 16S rRNA genes revealed that both cultures consisted Archaea belonging Sulfolobales. The gene clone library 4% (w/v) was dominated by unique phylotype related Sulfolobus shibatae (69% total clones). remaining clones affiliated Stygiolobus azoricus (11%), Metallosphaera sp. J1 (8%), Acidianus infernus (2%), novel Sulfurisphaera ohwakuensis (10%). In contrast, from MTC-B 12% did not appear contain shibatae-like organisms. Instead consortium (73.9% microorganisms detected similar those found in MTC-A.
