摘要: Abstract The present study describes systematic development of a gastroretentive bilayer tablet formulation containing fixed dose combination lamivudine and zidovudine exhibiting controlled drug release floating-bioadhesive characteristics. Effervescent tablets rational blend polymers were formulated by non-aqueous granulation employing isopropyl alcohol. A face centred cubic design was employed for optimizing various critical material attributes the tablets, which evaluated bioadhesive strength (BS), buoyancy time (T b ) characteristics as quality each layer. optimum composition layer selected “trading off” magnitude maximization BS, T characteristics, demarcated in space. Gamma scintigraphic studies healthy volunteers indicated behaviour optimized upto 6 h, while marketed immediate taken control showed maximum gastroretentivity only 1 h. In nutshell, these corroborated an optimal stable once-a-day system zidovudine.
