摘要: Publisher Summary This chapter discusses the DNA topoisomerases as anticancer drug targets. Much progress has been made in recent years understanding mechanism of action antitumor drugs that target topoisomerases. However, while it is now well established these interact with cleavable complex, molecular details protein-DNA-drug interaction are only just starting to emerge. The large body information available from study analogs provided some detailed on structural requirements for successfully complex. Little known about actual cell killing mechanism. question, which involves events beyond complex formation and its drugs, becoming more important. Knowledge signals involved killing, particular early induced by drug-mediated damage, might eventually lead discovery identification new targets drugs.
