Development of a DNA marker for variety discrimination specific to ‘Manten-Kirari’ based on an NGS-RNA sequence in Tartary buckwheat (Fagopyrum tataricum)

摘要: Abstract To discriminate the trace-rutinosidase variety of Tartary buckwheat ‘Manten-Kirari’, we developed DNA markers based on RNA polymorphism. Specifically, mapped 17.76 GB sequences, obtained using HiSeq2000, to create 11,358 large contigs constructed de novo from ‘Manten-Kirari’ derived GS-FLX+ titanium. From these, eight corresponding single- four-nucleotide polymorphisms between and ‘Hokkai T8’, which is representative normal rutinosidase content varieties in Japan. Using these markers, was discriminated T8’ by major three common buckwheats two markers. We also performed direct PCR flour dried noodle made with T8’. Based results, are promising for discriminating ‘Manten-Kirari’. This first study develop a marker Polygonaceae family including species.
