摘要: Abstract A 17-bp region between the −31 and −15 bp of mouse integrin αv gene is known to be one cis-acting elements for promoter activity. Experimental binding nuclear proteins −31/−15 reveals that −27/−16 mediates binding. The region, GGCTCCTCCTCC, has a TCCTCC motif, Sp1 motifs. An anti-Sp1 IgG an Sp1-binding oligonucleotide interfered with oligonucleotide, demonstrating binds region. In addition two other regions, −108/−89 −64/−44, were found bind within −108/+1 containing Ets-binding consensus sequence CAGGAAGT their binding, indicating both regions have functional site; which ACGGAAGT from −106 −99 ACTTCCTC −61 −54 bp, as deduced sequence. Mutations in or deletions any three elements, sites site, remarkably decreased activity detected Cotransfection Ets-1 cDNAs into B16F10 cells increased 2.9-fold.
