Longitudinal Assessment of Plasmodium falciparum var Gene Transcription in Naturally Infected Asymptomatic Children in Papua New Guinea

摘要: Sequestration and antigenic variation are essential for Plasmodium falciparum survival in vivo contributing to severe pathologic findings also chronic infection. Both conferred by P. erythrocyte membrane proteins encoded ~60 var genes. To study the dynamics of gene expression we conducted a 4-month longitudinal semi-immune children from Papua New Guinea. By use magnetic bead– anchored reverse-transcription polymerase chain reaction analysis performed over 5 regions as well cloning sequencing distribution full-length transcripts was analyzed. We identified dynamic picture with rapid switches but identical recurring up 10 weeks. The number correlated infections mean 1.7 per sample infecting strain. Analysis 158 different Duffy binding–like 1a sequences confirmed recombinogenic nature This is first report chronically infected children. (authors)
