摘要: Abstract Incorporation of the three synthetic hemins, Fe(III) meso-tetraalkylporphyrins with methyl, ethyl, or n-propyl groups, into apomyoglobin was followed by spectrophotometry, and stoichiometric complex formation confirmed. The reconstituted myoglobins bind an equimolar amount cyanide to exhibit visible absorption peaks at 419, 570, 608 nm. spectral feature independent concentrations. Proton NMR spectra complexes resolved pyrrole-proton signals hemins in a -5 -15-ppm region, which is comparable that corresponding deuterohemin-containing low-spin methemoproteins. These observations indicate presence NC-Fe-N(His-F8) structure presently metmyoglobins. metmyoglobins appeared as singlet, doublet, quartet for hemin complexes, respectively. systematic changes suggest dynamic free rotation alkylhemins about Fe-N(His-F8) bond. Temperature-dependent transition meso-tetraethylhemin-reconstituted myoglobin consistent thermally regulated heme pocket.
