摘要: The acquisition of mutations is relevant to every aspect genetics, including cancer and evolution species on Darwinian selection. Genome variations arise from rare stochastic imperfections cellular metabolism deficiencies in maintenance genes. Here, we established the genome-wide spectrum that accumulate a WT nine Saccharomyces cerevisiae mutator strains deficient for distinct genome processes: pol32Δ rad27Δ (replication), msh2Δ (mismatch repair), tsa1Δ (oxidative stress), mre11Δ (recombination), mec1Δ tel1Δ (DNA damage/S-phase checkpoints), pif1Δ (maintenance mitochondrial telomere length), cac1Δ cac3Δ (nucleosome deposition), clb5Δ (cell cycle progression). This study reveals diversity, complexity, ultimate unique nature each mutational spectrum, composed punctual mutations, chromosomal structural variations, and/or aneuploidies. produced clb5Δ/CCNB1, mec1Δ/ATR, tel1Δ/ATM, rad27Δ/FEN1 extensively reshape genome, following trajectory dependent previous events. It comprises transmission unstable genomes lead colony mosaicisms. comprehensive analytical approach defects provides model understand how might during clonal somatic cell populations, tumor cells.
