摘要: The use of the mammary gland transgenic goats as a bioreactor is well established platform for efficient production recombinant proteins, especially molecules that cannot be adequately produced in traditional systems using genetically engineered microorganisms and cells. However, extraordinary demand placed on secretory epithelium by expression large amounts protein, may result compromised physiology. In this study, milk composition was compared between control expressing high levels (1–5 g/l) human butyrylcholinesterase milk. Casein concentration, evaluated acid precipitation, significantly reduced with throughout lactation (P < 0.01). Milk fatty goats, determined gas chromatography, found to have fewer short chain acids 0.01) more saturated 0.05) controls, suggesting an overall metabolic stress and/or decreased key enzymes (e.g. synthase, stearoyl-CoA desaturase). concentration Na+, K+, assessed atomic absorption spectrophotometry, serum albumin, bromocresol green dye scanning densitometry, were similar during first several weeks lactation. progressed, significant increase Na albumin concentrations decrease K+ while animals remained unchanged These findings suggest that: (a) proteins associated slow-down other synthetic activities at epithelium, evidenced casein de-novo synthesis acids; (b) development permeable tight junctions main mechanism involved premature cessation secretion observed these goats.
