摘要: Tumor cell demise is an important event in the elimination of abnormal malignant cells and provides mechanism natural tumor suppression. Abnormalities incapacitating these finely tuned processes provide a strong advantage for cancer clones to succeed evading both physiological control systems therapeutic intervention. Expanding our knowledge molecular "crosstalks" that regulate crucial guiding successful design future anti-cancer therapeutics. Although currently available data indicate often depends on classical apoptotic pathways (mitochondrial and/or death-receptor pathways), evidence mounting alternative non-apoptotic may effectively contribute death. The assumption every organelle capable sensing, amplificating executing death also relatively novel unexplored concept. As recently shown, secretory pathway can be actively involved sensing stress stimuli possibly even initiating propagating signaling. Experimental indicates ER Golgi apparatus activate pro-survival (recovery) mechanisms as well suicide programs if stress-signaling threshold exceeded. It thus conceivable fragile balance protein trafficking between various subcellular compartments exceptional opportunity. Interestingly, growing number reports recognize targets, including proteins endoplasmic reticulum (ER) homeostasis. Further studies are, however, needed elucidate precise signaling emanating from ER-Golgi compartment.
