Regulation of Net Biosynthesis of Serum Albumin and Acute Phase Plasma Proteins INDUCTION OF ENHANCED NET SYNTHESIS OF FIBRINOGEN, α1-ACID GLYCOPROTEIN, α2 (ACUTE PHASE)-GLOBULIN, AND HAPTOGLOBIN BY AMINO ACIDS AND HORMONES DURING PERFUSION OF THE ISOLATED NORMAL RAT LIVER

摘要: Abstract Livers isolated from normal, male, fed rats were perfused for 12 hours with bovine or rabbit red blood cells suspended in Krebs-Ringer-bicarbonate solution containing serum albumin (3.0 g/100 ml) l-lysine-1-14C and 500 mg of glucose continuously infused. Net synthesis five specific plasma proteins was estimated by serological measurements changes their concentration perfusate. The influence insulin, cortisol, growth hormone, a nutritionally complete mixture amino acids on net evaluated. Control perfusions (no hormones added) resulted relatively linear the throughout experiments. In experiments full supplementation (insulin, acid mixture) fibrinogen haptoglobin increased 3-fold between 2 6 hours, α1-acid glycoprotein 3.5-fold 4 8 α2 (acute phase)-globulin 11-fold 10 hours. Increases latter four critically dependent upon presence cortisol represented first demonstration vitro hormone effects protein synthesis. These may be important understanding levels certain α β globulins that occur vivo mammals after many forms injury serve as useful model furthering our Maximum occurred positive nitrogen balance (more loss perfusate than urea gain). Under conditions employed combination insulin minimal partial required balance. Additional data are presented incorporation into total hepatic proteins, oxidation to 14CO2, production bile, utilization glucose, synthesis, nitrogen, defined liver perfusion system.
