摘要: Autophagy is a cell protective mechanism for maintaining cellular homeostasis. The present study aimed to investigate whether autophagy enhanced in the biomechanically induced degenerative cartilage of temporomandibular joint (TMJ) and potential role mitogen-activated protein kinase 3 (MAP4K3) mammalian Target rapamycin (mTOR) this observation. To induce changes TMJs, rats were subjected biomechanical dental stimulation by moving 4 molars away from their original position as we previously reported. ultrastructure autophagosome was observed transmission electron microscopy. number lysosomes analyzed flow cytometry. expression levels Beclin1 LC3 involvement MAP4K3 activity detected immunohistochemistry, real-time PCR western blot. mTOR pathway indicated p-mTOR p-p70S6 K assayed TMJ degeneration, characterized irregular arrangement cell-free area, experimental groups. Under microscopy, presence autophagosomes, small patches condensed chromatin, abundant rough endoplasmic reticulum Golgi apparatus. increased, while level decreased Cartilage which be exhibited accompanied reduced activity.
