摘要: Partially inbred, MHC homozygous miniature swine have been used to study the nature of human xenogeneic cellular immune response Ags in vitro. Human T cells responded xeno-MHC MLR at least as well they did allo-MHC and appeared share similar requirements for APC either stimulator (direct pathway) or responder (indirect derivation. In addition, mAb-blocking experiments indicated that majority primary anti-pig xeno-response was directed toward porcine class II involved interaction with CD4 accessory molecule. Finally, availability intra-MHC recombinant haplotypes our herds has made it possible map genetically antigenic specificities recognized anti-swine responses. For this purpose, cell clones were generated from cultures tested reactivity haplotypes. Clear evidence antixenogeneic observed. all cases which allelic differences between detected (the clones), could be mapped region cross-reactivity consistent known sequence similarities DR beta-chains. Our results indicate, therefore, anti-porcine is strength specificity an allogeneic response, TCR repertoire, molecule interactions, cytokine production required both direct indirect pathways recognition responses are functionally intact.
