摘要: Summary Purpose: Increased interictal concentrations of extracellular hippocampal glutamate have been implicated in the pathophysiology temporal lobe epilepsy (TLE). Recent studies suggest that perturbations metabolizing enzymes glutamine synthetase (GS) and phosphate activated glutaminase (PAG) may underlie excess TLE. However, molecular mechanism enzyme remains unclear. A better understanding regulatory mechanisms GS PAG could facilitate discovery novel therapeutics for TLE. Methods: We used situ hybridization on histologic sections to assess distribution quantity messenger RNA (mRNA) subfields formations from following: (1) patients with TLE concomitant sclerosis, (2) no (3) nonepilepsy autopsy subjects. Key Findings: mRNA was increased by ∼50% CA3 without sclerosis versus subjects. >100% subiculum both patient categories also CA1, CA2, CA3, dentate hilus sclerosis. Finally, gyrus subjects, sclerosis. Significance: These findings demonstrate complex changes expression mRNAs formation TLE, raise possibility transcriptional posttranscriptional regulation proteins epileptic brain.
