摘要: We report here a polymerase chain reaction (PCR)-based DNA profiling technique that permits Trypanosoma cruzi strain characterization by direct study of infected tissues. This is based on application recently developed method fragment identification, called low-stringency single specific primer PCR (LSSP-PCR), to the variable region kinetoplast (kDNA) minicircles from T. Thus, we can translate intraspecific polymorphism in nucleotide sequence kDNA into and highly reproducible signature. Comparison with phenogram obtained fingerprinting analysis set strains showed good qualitative correlation between degree divergence LSSP-PCR profiles genetic distance strains. signatures heart tissue acutely or chronically animals revealed perfect concordance patterns cultured parasites for CL Colombiana but not Y strain, which known be multiclonal. However, match was studies two clones strain. take this as evidence some multiclonal there heterogeneity among tropism tissue. Finally, it possible obtain signature human patient chronic Chagasic myocardiopathy. sequences amplified tissues constitute new tool molecular epidemiology Chagas' disease.
