摘要: Publisher Summary This chapter discusses the methods for studying polymerase translocation. In model translocation can slide on template over a range that restricts it to primarily occupy two positions: pre-translocated position, in which 3´-nt of RNA occupies site, is bound by substrate NTP during catalysis, and post-translocated has cleared binding site. been tested both developing evaluating its kinetic implications, monitoring position transcript elongation with exonucleases. Kinetic effects rarely allow unambiguous interpretation mechanism, therefore important use direct probe test models mechanism. Exonucleases may be more than area foot printing reagents this respect because single nucleotide shift diffuse footprint extends 20 or 30 nucleotides difficult observe, while similar at digestion an exonuclease blocked readily detected.
